Sterile handling
Contamination is the failure mode that does not announce itself. A cloudy vial is obvious; a lightly contaminated one simply produces results that will not replicate, and the cause is usually attributed to something else entirely.
Before opening anything
- Work on a cleared surface that has been wiped with 70% isopropyl alcohol and allowed to dry. Alcohol needs contact time; wiping and immediately proceeding does very little.
- Assemble everything first. Opening a vial and then hunting for a syringe leaves the septum exposed for longer than necessary.
- Wipe the vial septum with a fresh alcohol pad and let it dry before the needle goes anywhere near it. A wet septum carries whatever the pad picked up straight into the vial.
While working
- One needle, one entry. Re-entering a vial with a needle that has touched any other surface is the single most common route to contamination.
- Keep the vial upright and avoid touching the septum with anything except the needle.
- Do not flame, boil or autoclave a peptide vial. Heat destroys the material you are trying to preserve.
Documentation
Label every reconstituted vial at the moment it is prepared, not afterwards. Record the compound, the mass taken into solution, the volume of solvent, the resulting concentration, the date, and the lot number from the original vial. An unlabelled vial in a freezer is waste, because nothing in it can be trusted or reported.
Traceability back to a lot number is what separates a documented result from an anecdote. If the vial cannot be traced to a certificate, the data it produced cannot be defended.
